Patients with B-cell malignancies and autoimmune disorders often face relapse due to antigen loss following CD19-CAR T cell therapy. The lack of alternative tumor targets contributes to disease recurrence and limits treatment options for these patients.
Solution
PeproMene Bio is developing BAFF-R CAR-T therapy (PMB-CT01), an immunotherapy that targets the B-cell Activating Factor Receptor (BAFF-R) to address B-cell malignancies and autoimmune disorders. Unlike CD19-CAR T cell therapies, BAFF-R targeting is less susceptible to antigen loss-related relapse, offering a more durable response. The therapy involves genetically engineering a patient's T cells to express chimeric antigen receptors (CARs) that recognize and bind to BAFF-R on tumor cells. These modified T cells are then infused back into the patient to selectively target and eliminate BAFF-R-expressing malignant B cells. PeproMene Bio is also developing BAFF-R/CD3 bispecific antibodies (BiTEs) and allogeneic BAFFR CAR-NK cell therapies.
