Problem
Glioblastoma (GBM) is the most common and aggressive primary brain tumor in adults, progressing quickly and resisting standard treatments, with fewer than 7% of patients surviving five years post-diagnosis. No therapy has meaningfully extended survival for patients whose disease returns after initial treatment, and conventional cell therapies have been ineffective against solid tumors due to heterogeneity, antigen escape, and T cell exhaustion.
Solution
Altido Therapeutics offers CAR-TEAM, a dual-mechanism immunotherapy platform engineered to overcome barriers that have limited cell therapies in solid tumors. Each dose uses a patient's own T cells, genetically modified to express a chimeric antigen receptor (CAR) targeting one tumor antigen while simultaneously secreting bispecific T-cell-engaging antibody molecules (TEAMs) that direct both CAR-T and bystander T cells against a second antigen. This two-pronged attack addresses tumor heterogeneity and antigen escape, while the design aims to reduce T cell exhaustion. The lead program, ALTX-100, targets EGFRvIII and EGFR in glioblastoma and is administered intraventricularly—directly into the cerebrospinal fluid—to bypass the blood-brain barrier and concentrate therapy at the tumor site.
